On my street I have a doctors surgery and outside on the public road is a parking bay with the road marking 'Ambulances' and this sign.

Does this mean I can park in the ambulance bay on Saturdays and Sundays without any risk of a PCN? Any clarification would be much appreciated.

Yes - does not apply on Sat-Sun. But there should be a single yellow line too.

Show the Google Street View location.

Ok, doesn't need a line I think but the bay markings should be in yellow?

In any case you are fine to park there outside of the restricted hours.

I wouldn't like to see a fire engine try to get down that road on a weekend if vehicles are parked on both sides.
Looks very narrow.

The road is part of Reading Borough Council's residents parking zones.

Signage is IMO all to pot.
If No Waiting with a yellow plate, should have a narrow yellow line and the bay.
Or a bay and a white plate with parking for Ambulances only during restricted times.
Whether an adjudicator would agree is a different matter.

That's what I thought at first but signs manual gives the example top right (could be ambulances or taxis etc). But the bay outline should be yellow as well as the words.

Yellow line not needed because there is no variance between restriction on ambulance/taxi parking and bay parking generally. I think.

Hmmm
TSRGD 2016 is a little reticent on with single yellow or not.
And is 2002 version.
But both say yellow box.
However can hear an adjudicator saying substantially compliant.

That's my feeling also The only point would be why would you look outside a bay with no yellow line

I would suggest 1028.6 should have been used (schedule 7 part 4 item 8)

www.legislation.gov.uk/uksi/2016/362/schedule/7/made
